Drug intelligence / Profile preview
[161Tb]Tb-DOTA-N4MU01 is a preclinical-stage targeted radioligand therapy (radioimmunoconjugate) consisting of the fully human anti-Nectin-4 monoclonal antibody N4MU01 conjugated to the macrocyclic chelator DOTA and radiolabeled with the beta- and Auger-electron-emitting radioisotope terbium-161 ([161Tb]). Developed by researchers at the University of Saskatchewan and Université Laval, the agent specifically targets Nectin-4, a cell adhesion molecule overexpressed in various cancers, including triple-negative breast cancer (TNBC) and non-small cell lung cancer (NSCLC). Upon binding to Nectin-4, the radioimmunoconjugate is internalized, delivering localized radiation that induces DNA damage and tumor cell death. Preclinical studies have demonstrated significant anti-tumor efficacy and survival benefits, particularly when combined with immune checkpoint inhibitors like anti-PD-L1 antibodies.
